Keeping ITRs intact for best AAV performance

PlasmidFactory’s proprietary ITRPROTECT®and ITRRESCUE® technologies enables the stable amplification of plasmid DNA containing sensitive inverted terminal repeats (ITRs) — a critical requirement for achieving highest AAV vector performance.
Traditional quality control methods such as SmaI restriction analysis cannot reliably quantify the fraction of non-intact ITRs. ITRPROTECT® combines an optimized fermentation strategy with advanced Next Generation Sequencing (NGS) analytics to verify ITR integrity, accurately quantify intact versus rearranged sequences, and ensure stable amplification of full-length ITRs.
Beyond analysis, ITRRESCUE® enables the restoration of defective ITR regions — allowing replacement or repair of truncated or rearranged ITRs to re-establish full-length functional ITR sequences.
GFP reporter gene cassettes flanked by intact ITR2 sequences are available in stock as plasmid DNA or Minicircle DNA, including both single-stranded (ss) and self-complementary (sc) configurations.
