AAV packaging and helper constructs for high-efficiency vector production

PlasmidFactory provides plasmid and Minicircle DNA specifically designed for Adeno Associated Virus (AAV) production workflows, including AAV transfer plasmids as well as packaging and pHelper plasmids, the latter two combined on a single construct to provide essential components for efficient virus packaging systems. These components support efficient AAV assembly and reliable transgene expression in research and (pre)clinical settings.
Our vectors enable coordinated rep/cap expression, genome delivery, and adenoviral helper functions, ensuring robust AAV replication and packaging without the need for a helper virus. Well-defined ITR sequences ensure efficient packaging within the typical AAV capacity and enable reliable transduction across multiple AAV serotypes and capsids.
Minicircle DNA can further enhance AAV production workflows by eliminating bacterial backbone sequences, reducing unwanted DNA packaging (“back-packing”) and thereby improving purity, efficiency and safety. These advantages can contribute to cleaner viral vector preparations, better packaging, higher titers and increased transduction efficiency (learn more: AAV vector production).
PlasmidFactory holds the exclusive worldwide license from the German Cancer Research Center (DKFZ) for the 2-plasmid system used in AAV vector production, including the pDG and pDP plasmid families. These plasmids are widely used to support efficient and scalable AAV manufacturing.

Preserving ITR Integrity for Reliable AAV Production
PlasmidFactory’s proprietary ITRRESCUE® and ITRPROTECT® technologies support the reliable amplification of AAV transfer constructs containing sensitive inverted terminal repeats (ITRs), helping to maintain or restore intact sequences required for efficient AAV yield, purity, and packaging.
Through optimized production processes and next-generation sequencing analysis, ITR-containing AAV constructs can be amplified without ITR degradation using ITRPROTECT®, ITR integrity can be verified, and truncated or damaged ITRs can be restored using ITRRESCUE®.
Our AAV in-stock constructs include transfer vectors with ITR-flanked GFP reporter cassettes for single-stranded (ssAAV) and self-complementary (scAAV) applications. These constructs apply our ITR technologies, are designed to contain fully intact ITR sequences, and are available as plasmid or Minicircle DNA.
ITRPTROTECT® and ITRRESCUE® can also be applied to customer-specific plasmid and Minicircle constructs to support stable amplification of ITR-containing vectors.
